BrdU标记的同种异体骨髓基质干细胞移植治疗兔股骨头坏死骨缺损模型Experimental study on osteonecrosis and bone defect of rabbit femoral head model treated with BrdU-marked allogeneic bone marrow stromal cells

摘要(Abstract):
[目的]探讨同种异体骨髓基质干细胞移植治疗股骨头坏死的可行性,并研究移植细胞的转归和缺损区新生骨的来源。[方法]24只新西兰大耳白兔随机分成两组,每组12只,所有动物的双侧股骨头均参加实验。用液氮冷冻法造模,股骨头钻孔造成骨缺损。对照组置入胶原蛋白海绵,实验组置入复合有定向诱导并用BrdU标记的第5代同种异体骨髓基质干细胞的胶原蛋白海绵。分别于术后2、4、8、12周处死3只实验动物。所获的股骨头标本分别做组织切片HE染色、BrdU免疫组织化学染色,进一步测算股骨头冠状截面的新生骨小梁在骨缺损区所占的面积百分比,做统计分析。[结果]股骨头标本切片后行免疫组织化学染色,可见成骨细胞及骨细胞的核均呈棕褐色。实验组缺损区2周时新生骨小梁开始形成,并可见少量的浆细胞和淋巴细胞,4周时浆细胞和淋巴细胞明显减少,12周时缺损区可见大量细小的骨小梁,炎性细胞消失。两组实验动物股骨头冠状截面的新生骨小梁在骨缺损区所占的面积百分比具有统计学意义上的差异(P<0.05)。[结论]移植的骨髓基质干细胞的最终转归是缺损区新生骨小梁,同种异体骨髓基质干细胞可以用于股骨头坏死的治疗。
