经尾静脉移植干细胞治疗神经Crush损伤的研究Effects of intravenous transplanted stem cells on nerve crush injury

摘要(Abstract):
[目的]探讨经尾静脉移植骨髓间充质干细胞对SD大鼠周围神经Crush损伤的修复作用。[方法]建立SD大鼠5 mm坐骨神经Crush损伤模型,然后将实验动物分为两组:干细胞治疗组和对照组(生理盐水组)。Crush损伤模型建立后1周,经尾静脉分别注射5×106干细胞(干细胞治疗组)或生理盐水(生理盐水组),然后于不同的时间点分别借助于活体荧光成像系统、坐骨神经指数测量、电生理检测和神经肌肉病理学等方法评价干细胞的归巢现象及其对神经Crush损伤的修复作用。[结果]活体荧光成像系统检测到干细胞治疗组的损伤侧神经Wallerian变性区域有高强度荧光信号聚集;SFI测量证实在损伤后2、4、6周三个不同时间点,干细胞治疗组SFI的恢复优于对照组;同样的,电生理检测和神经肌肉病理学均提示干细胞治疗组的促神经损伤恢复效果优于对照组。[结论]经尾静脉移植骨髓间充质干细胞可以促进周围神经Crush损伤的修复。
