基于永生化表皮细胞工程化皮瓣体外形成的实验研究Experimental study of engineering flap formation based on immortalized keratinocytes in vitro

摘要(Abstract):
目的:探讨永生化上皮细胞在工程化皮瓣中应用的可能性。方法:把人端粒酶催化亚基(hTERT)转染兔表皮细胞,筛选阳性克隆并在体外进行大量扩增;将扩增的永生化表皮细胞和平滑肌细胞与支架(PGA)复合并在体外培育后,植入裸鼠皮下;3~6个月取材,进行形态学和免疫组织化学评价。结果:实验组,术后6个月,复合体被新生的表皮和平滑肌组织所覆盖,抗人角细胞广谱角蛋白(AE1/AE3)抗体和抗人平滑肌a-肌动蛋白抗体染色呈强阳性,与对照组差别有显著性意义(P<0.01)。结论:永生化表皮细胞在皮瓣组织工程中具有较好的应用前景。
