细胞凋亡在失神经骨折愈合过程中的作用机制The role of apoptosis during the process of fracture healing in a denervation state

摘要(Abstract):
[目的]通过观察失神经后大鼠胫骨骨折愈合情况,探讨细胞凋亡在失神经骨折愈合过程中发生的规律及意义。[方法]将96只成年健康雌性SD大鼠随机分为实验组和对照组。实验组建立T10脊髓完全性损伤状态下大鼠胫骨骨折模型,对照组行单纯胫骨骨折术,分别于术后3、5、7、14、21及28 d取材(每个时间点n=8),于骨折断端上下各5 mm处剪断胫骨,经脱钙及固定后,制成6μm切片。切片应用TUNEL法原位观察细胞凋亡的时间分布。另外还包括动物行为学观察以及术后7、14、21和28 d拍摄X线片。[结果]在骨折愈合过程中,两组均可见凋亡细胞,实验组的凋亡指数在术后各时间点均高于对照组,差异具有统计学意义(P<0.05),并具有时间分布的特点,两组均以术后7 d出现高峰,随着时间的延长,实验组和对照组的细胞凋亡指数逐渐减小。[结论]细胞凋亡是脊髓损伤后重要的病理变化,机体通过凋亡机制控制细胞数量并清除无用的细胞,以利于失神经状态下的骨折修复愈合。
