三维动态灌注法构建大段组织工程化骨修复羊胫骨干节段性缺损Repair of experimental long bone defect with large tissue engineered bone created by 3-D perfusion culture

摘要(Abstract):
[目的]观察三维动态灌注法构建的大段组织工程化骨修复羊胫骨干节段性骨缺损的效果。[方法]利用自行设计的灌注型生物反应器对复合骨髓基质干细胞的大段磷酸三钙柱状载体进行动态灌注培养或静态培养2周,通过葡萄糖日耗量、乳酸生成量监测细胞在载体内的扩增。在羊胫骨干节段性骨缺损中分别植入β磷酸三钙、静态培养和动态灌注培养的组织工程化骨,通过X线片、大体观察、硬组织切片和形态计量学检查观察其修复骨缺损的效果。[结果]动态培养法葡萄糖日耗量及乳酸生成量明显高于静态培养法。单纯β磷酸三钙、静态培养和动态灌注培养的组织工程化骨植入骨缺损16周后的临床愈合率分别为0/6、2/6、4/6。动态灌注培养组的材料残余率明显低于单存材料组(P<0.05)。[结论]三维动态灌注培养法构建的大段组织工程化骨修复羊长骨节段性缺损的效果优于传统静态培养法。
