骨组织工程支架A-W-MGC/CS的体内成骨实验研究Biocompatibility and in vivo osteogenic capability of novel bone tissue engineering scaffold A-W-MGC∕CS

摘要(Abstract):
[目的]探讨多孔纳米磷灰石-硅灰石生物活性玻璃陶瓷/壳聚糖复合支架材料(A-W-MGC∕CS)载体承载粘附腺病毒载体的种子细胞体内成骨能力。[方法]培养兔骨髓间充质干细胞(BMSCs),转染Ad-GFPh BMP后接种于载体上,电镜观察贴附情况;MTT检测增殖情况;制备日本大耳白兔桡骨1.5 cm骨缺损模型,分三组。实验组1植入经Ad-GFP-h BMP转染的BMSCs载体;实验组2仅植入载体;空白组不植入任何载体,仅注射经Ad-GFP-h BMP转染的BMSCs。术后12周X线片观察骨缺损修复情况,取出缺损区标本行病理切片观察。[结果]A-W-MGC∕CS载体可较好贴附BMSCs且对细胞增殖无明显影响。实验组1骨缺损修复较好,支架材料完全降解;实验组2新生骨质量较差;空白组未见缺损修复,主要由纤维结缔组织填充。[结论]A-W-MGC∕CS构建的组织工程骨具有较好的骨缺损修复能力。
