依降钙素对骨质疏松-骨关节炎大鼠的影响Effect of elcatonin on osteoporosis and osteoarthritis in rats

摘要(Abstract):
[目的]观察不同剂量依降钙素注射治疗对骨质疏松-骨关节炎(OP-OA)模型大鼠的软骨下骨的影响。[方法] 3个月龄雌性SD大鼠32只随机分为4组;除空白组外,均采用双侧前交叉韧带切断和卵巢切除方法建立OP-OA模型。低剂量组和高剂量组分别肌肉注射依降钙素5 IU/kg和10 IU/kg,1次/周。空白组和模型组仅注射生理盐水。术后12周处死动物,检测血清骨代谢指标,行组织学评分和显微CT骨定量分析。[结果]依降钙素两组CTX-Ⅰ、CTX-Ⅱ、IL-1、IL-6、TRACP 5b含量均较模型组降低(P<0.05),且高剂量组更明显。依降钙素两组OARSI评级低于模型组,但差异无统计学意义(P>0.05)。与模型组比较,依降钙素两组BV/TV、Tb.N显著增高(P<0.05),而Tb.Sp显著降低(P<0.05),其中高剂量组的效果优于低剂量组(P<0.05)。[结论]依降钙素能改善骨关节炎模型大鼠骨代谢活性及预防软骨下骨骨质疏松,且呈剂量依赖性。
