α-半水硫酸钙/辛伐他汀/万古霉素复合材料抗菌和细胞毒性作用的研究Antibacterial properties and cytotoxicity of alpha half water calcium sulfate/simvastatin/vancomycin composite materials

摘要(Abstract):
[目的]研究不同浓度配比α-半水硫酸钙/辛伐他汀/万古霉素复合材料的体外抗金黄色葡萄球菌、大肠埃希菌和铜绿假单胞菌的作用,以及α-半水硫酸钙/辛伐他汀/万古霉素复合材料浸提液对新生小鼠成骨细胞的毒性作用。[方法]采用细菌生长抑制的定性和定量试验测定抑菌圈直径、抑菌率;MTT法细胞毒性试验测定α-半水硫酸钙/辛伐他汀/万古霉素复合材料浸提液体外对成骨细胞的相对增殖率及观察细胞形态学改变,并确定相应毒性等级。[结果]含5、10、20μg/mlα-半水硫酸钙/辛伐他汀/万古霉素复合材料对新生小鼠成骨细胞作用呈浓度-效应关系,对金黄色葡萄球菌和大肠埃希菌的抑菌圈平均直径分别为28、30、33mm和24、23、25mm,对铜绿假单胞菌没有抑菌作用。[结论]α-半水硫酸钙/辛伐他汀/万古霉素复合材料浸提液基本无细胞毒性,细胞相容性好,具有良好的抗菌效果,并能促进成骨细胞增殖和分化。
